KARMAC-001 is a CAR-engineered monocyte targeting TREM2, a scavenger receptor highly expressed on immunosuppressive TAMs, with limited expression in tissue-resident macrophages. TREM2+ TAMs are key drivers of immunosuppression across solid tumors. Leveraging their innate chemokine-driven homing, infused TREM2-CAR-monocytes traffic to tumors and differentiate into macrophages capable of eliminating TREM2+ TAMs, thereby disrupting the immunosuppressive tumor microenvironment.
